摘 要:目的探讨大鼠实验性隐睾对睾丸间质细胞数量、形态及相关功能的影响。方法采用手术诱导方法制作大鼠(6月龄)双侧隐睾模型(每组6只),分别于术后第3天、7天、20天和28天取左侧睾丸入Bouin液固定,假手术组作为对照组。应用体视学方法检测睾丸间质细胞的数量、形态变化;应用免疫组织化学检测细胞分裂周期蛋白25A(CDC25A)、干细胞因子(SCF)、雄激素受体(AR)和雌激素受体α(ERα)的表达变化。结果隐睾术后睾丸间质体积分数明显增加,但睾丸间质的绝对体积无明显变化。间质细胞数量和光镜形态也无明显变化。免疫组织化学方法显示,CDC25A和SCF在间质细胞区域强表达,隐睾对其表达无明显影响;AR在间质细胞表达较弱,位于细胞质内,各组之间表达强度无明显变化;ERα在对照组睾丸间质细胞内表达较弱,间质细胞核基本无阳性着色,但随着隐睾时间的延长,间质细胞核阳性表达比例明显升高,在隐睾术后第28天时达到93.6%。结论大鼠实验性隐睾对睾丸间质细胞的数量和光镜形态无明显影响,但对其甾体类激素合成及相关调节功能有明显影响。Objective To investigate the change of the number, morphology and function of Leydig cells after induction of eryptorchidism. Method Adult rats were induced cryptorchidism by confining the testes of both sides into the abdominal cavity. They were decapitated at intervals of 3, 7, 20 and 28 days postoperation (6 rats in each group). Sham group as the control. The number and morphological changes of Leydig ceils were detected by the method of stereology. Application of immunohistochemistry detected of the expression of cell division cycle 25 A (CDC25A) , stem cell factor (SCF) , androgen receptor (AR) , estrogen receptor o~ (ERct)in Lcydig cells. Result The volume fraction of testieular interstitium increased significantly, but the absolute volume of testicular interstitium unchanged after induction of cryptorchidism. The number and morphology of Leydig ceils did not change significantly. Immunohistochemical staining showed CDC25A, SCF strongly expressed in the area of Leydig cells, and the cryptorchidism had no significant effect on their expression. AR had weak staining in the cytoplasm of Leydig cells, and the intensity of AR expression no significant changes between different groups. The expression of ERα in Leydig cells in the control group was weak, almost no positive nuclei. But the percentage of Leydig cells which has ERα positive nucleus increased significantly with the prolongation of cryptorchidism time. It had reached 93.6% after 28 days cryptorchidism. Conclusion Experimental cryptorchidism has no significant effect on the number and light microscope morphology of Leydig cells, but it has significant effect on the function of steroid hormone synthesis.
