摘 要:目的探讨自噬调节对胶质瘤起始细胞辐射敏感性的影响。方法收集手术切除的胶质瘤标本,制备胶质瘤起始细胞系SU-2,用雷帕霉素处理后再行X线照射(A组)或直接行X线照射(B组)。MTT比色法和克隆形成实验观察SU-2细胞增殖情况;应用Hoechst 33258荧光染色检测SU-2细胞凋亡。结果 A组的细胞增殖率和克隆形成率较B组明显降低(P<0.01),细胞凋亡率显著增加。结论胶质瘤起始细胞经自噬激动剂雷帕霉素处理后,对X线照射的辐射敏感性明显增加。Objective To investigate the effects of rapamycin on the sensitivity of glioma-initiating cells(GICs) to radiation.Methods Glioblastoma samples from surgery were collected and glioma-initiating cells(SU-2) were prepared.The SU-2 cells were treated with rapamycin,which was followed by radiation of X-ray in group A.The SU-2 cells in group B were given radiation of X-ray alone.The growth of SU-2 cells was assessed with MTT method and cell apoptosis was detected by Hoechst 33258 fluorescence staining.Results The proliferation rate and the number of colonies were significantly lower in group A than those in group B(P0.01).The cell apoptosis was significantly higher in group A than that in group B.Conclusion The sensitivity of GICs to radiation is remarkably increased by pretreatment with rapamycin.
