摘　　要：目的对大鼠胃溃疡模型制备方法进行改进。方法胃粘膜表面局部投予乙酸制备大鼠胃溃疡模型。10%水合氯醛(3.5ml/kg体重)肌肉注射麻醉,用0.5%碘伏消毒大鼠腹部皮肤,沿左肋下缘开腹,将胃轻轻拉出。用带有圆环的胃溃疡制备镊子夹住距幽门部3mm胃体部区域。在夹闭区的胃腔内注入0.18ml矿物油和0.02ml60%乙酸,45sec后从胃部吸出乙酸,再向胃腔内注入2ml生理盐水,1min后吸出。将胃复位,逐层关腹。结果该方法可在大鼠胃部造成单个、圆形、大小、深度一致的溃疡灶,并且解决了手术引起粘连的问题。结论本文提供一种改进的胃粘膜表面局部投与乙酸制备大鼠胃溃疡模型的方法,该模型可用于考察消化性溃疡的愈合过程、抗溃疡药物疗效评价、药物对胃粘膜所产生的副作用监测等。Objective To improve the established method of acetic acid-induced rat gastric ulcer model. Methods The rat gastric ulcer model by applying acetic acid to specified area in rat＇s gastric mucosa. After 10% chloral hydrate intramuscular anesthesia （3.5ml/kg）, the rat＇s skin of abdomen was sterilized by 0.5% iodophors. The rat＇s abdomen was opened along left subcostal margin and the stomach was gently pulled out. The body of stomach approximately 3 mm proximal to the pylorus was clamped by round forceps. Then a mixture of mineral oil（0.18 ml） as well as 60% acetic acid（0.02 ml） were injected into the clamped lumen, which was removed after 45 seconeds. After that, normal saline（2ml） was injected into the gastral cavity, which was removed after 1 minute. Finally, the abdomen was closed. Results A clearly defined deep round ulcer was induced, which was free of post-operative adhesion.Conclusion A kind of improved acetic acid-induced rat gastric ulcer model was established.It could be used to assess the healing process of peptic ulcer, therapeutic effect of antiulcer drug and side effect to gastric mucosa which was caused by drugs.
