摘 要:目的采用细胞培养法在体外评估镁合金表面不同硅含量的羟基磷酸钙涂层对成骨细胞的毒性反应。方法采用经鉴定的SD大鼠成骨细胞接种于三种含有不同浓度硅含量的羟基磷酸钙涂层的镁合金及对照组裸镁合金,MTT法检测细胞活力并检测成骨细胞的碱性磷酸酶表达;扫描电镜观察细胞生长情况。结果细胞活力检测和碱性磷酸酶活性分析均表明,硅含量在5%及10%的羟基磷酸钙涂层利于成骨细胞在镁合金表面的生长;扫描电镜下可见,成骨细胞在5%及10%硅含量的涂层上呈聚集性生长且分布均匀,而在裸镁合金上生长缓慢且稀疏分布。结论镁合金表面硅含量为5%及10%的羟基磷酸钙涂层都利于成骨细胞的体外黏附和生长。Objective To evaluate the cytotoxicreaction of calcium hydroxyapatite coating with different silicon concentration of Mg-Mn-Zn-Ca. Methods The identified osteoblasts of SD rats were seeded on calcium hydroxyapatite coating with three kinds of different silicon concentration of Mg-Mn-Zn-Ca and naked ones. The cell vitality was detected with MTT assay, and alkaline phosphatase(ALP) vitality was measured by ALP detection kit. The growth of osteoblasts on different concentrations of coatings and naked magnesium were observed with scanning electron microscopy(SEM). Results Both MTT assay and ALP analysis showed that hydroxyapatite coating with 5%~10% concentrations of silicon in weight was able to promote the growth of osteoblasts on magnesium alloy. SEM displayed that osteoblasts on hydroxyapatite coating with 5%~10% concentrations of silicon in weight aggregated gradually and were well-distributed, while cells on naked ones growed slowly and present sparse distribution. Conclusion Hydroxyapatite coating with 5%~10% concentrations of silicon in weight on Mg-Mn-Zn-Ca promotes the growth, adhension and proliferation of osteoblasts in vitro.
