摘  要:目的 探讨香烟提取物(CSE)对人支气管上皮细胞表达人β防御素2(hBD2)的影响。方法利用不同浓度(4%和8%)的CSE刺激人支气管上皮细胞(16HBE),分别于0、6、12、24、48 h收集细胞总蛋白,运用酶联免疫吸附法(ELISA)测定hBD2的蛋白表达水平。结果 4%和8%CSE均诱导16HBE于6、12、24、48 h进行性hBD2表达下降。8%CSE干预组变化幅度大于4%CSE干预组。结论 CSE抑制人支气管上皮细胞表达hBD2,此机制可能参与香烟对气道防御的抑制作用。Objective To investigate the effect of cigarette smoke extract(CSE)on human β-defensin 2(hBD2)expression by human bronchial epithelial cell(16HBE).Methods 16HBE cells were incubated with two different concentrations of CSE(4% and 8%),and the cell protein was harvested at the experimental timepoints of 0 h,6 h,12 h,24 h,48 h,respectively.ELISA was used to assess the hBD2 protein expression level.Results After CSE(4% and 8%)stimulation,the hBD2 expression level significantly decreased at 6 h,12 h,24 h,48 h timepoints time-dependently,compared with the control.The expression of hBD2 in CSE 8% group had a larger change extent than CSE 4% group.Conclusion CSE suppresses hBD2 expression by 16HBE cells,and this mechanism may contribute to the inhibition of airway defense by cigarette smoke.
