摘  要:目的观察复方天葡片(cTP)对辐射损伤小鼠免疫与造血功能恢复的促进作用。方法 6周龄清洁级雄性ICR小鼠108只,随机分为正常对照组、模型组、阳性对照组及cTP高、中、低剂量组,每组18只。cTP高、中、低剂量组动物分别给予0.06、0.12、0.24g/kg cTP灌胃,阳性对照组给予20mg/kg利可君灌胃,正常对照组及模型对照组用等体积蒸馏水灌胃,均为1次/d,连续21d。然后采用3.0Gy60Coγ放射源照射除正常对照组外的动物建立辐射损伤模型,并继续灌胃给药10d至实验结束。测定各组小鼠的淋巴细胞转化率、巨噬细胞吞噬作用、外周血白细胞数及红系(CFU-E、BFU-E)、粒-单核细胞系(CFU-GM)、巨核系(CFU-Meg)、混合系(CFU-Mix)集落生成单位并进行组间比较。结果与正常对照组比较,模型组小鼠经3.0Gy60Coγ照射后,T淋巴细胞、B淋巴细胞转化功能以及巨噬细胞吞噬功能均明显下降(P<0.01),而cTP可明显提高辐射损伤后小鼠淋巴细胞转化率及巨噬细胞吞噬作用;此外,与正常对照组比较,模型组小鼠接受3.0Gy60Coγ射线照射后外周血白细胞数量迅速降低(P<0.01),造血系统受损明显,CFU-E、BFU-E、CFU-GM、CFU-MEG和CFU-MIX数量急剧下降(P<0.001),而cTP可促进白细胞恢复,粒细胞-单核细胞集落、红细胞系集落生成单位、混合系集落生成单位生成数增加。结论 cTP具有明显的辐射防护作用,其作用机制可能与提高机体免疫功能及造血功能有关。Objective To observe the effect of comlex TIANPU tablets(cTP) on the immune and hematopoietic function of radiation-injured mice.Methods One hundred and eight male ICR mice(aged 6 weeks) were involved in present study and randomly divided into normal control group,model control group,positive control group,and cTP high-,middle-and low-dose group(18 each).Mice in cTP high-,middle-and low-dose group were administratered intragastrically with 0.24,0.12 and 0.06g/kg cTP,respectively;in positive control group with 20mg/kg leucogen;while in normal and model control group with same amount of distilled water.All the animals were administratered once a day for 21d.The radiation injury model was then established by radiation of 3.0 Gy ^60Co γ rays in all mice except those in normal control group,and the intragastric administration was continued for 10 days after radiation(till the end of the experiment).The lymphocyte transformation,phagocytosis of phagocytes and the peripheral leukocyte count were determined,and the colony forming unit-erythroid(CFU-E),burst forming unit-erythroid(BFU-E),CFU-granulocyte and macrophage(CFU-GM),CFU-megakaryocyte(CFU-Meg) and CFU-multipotential(CFU-Mix) in bone marrow were tested.Results After 3.0Gy ^60Co γ rays radiation,the T and B lymphocyte transformation and phagocytosis of phagocytes of model control group decreased significantly compared with those of normal control group(P〈0.01),while cTP increased the lymphocyte transformation and phagocytosis of phagocytes of model mice.Moreover,compared with normal control group,the CFU-E,BFU-E,CFU-GM,CFU-Meg and CFU-Mix in bone marrow and peripheral leukocyte count of model control group decreased rapidly after 3.0Gy ^60Co γ ray radiation(P〈0.01),while cTP promoted the recovery of leukocytes and increased the CFU-E,BFU-E,CFU-GM,CFU-Meg and CFU-Mix.Conclusion cTP has a forceful effect against the radiation-induced damage in a possible mechanism of enhancing the immune and hematopoietic function.
