摘  要:目的:探讨透骨消痛颗粒对兔膝骨性关节炎软骨及软骨下骨的影响。方法:取新西兰大白兔36只,雌雄各半,根据改良Hulth法制作骨性关节炎模型,动物随机分成正常对照组、骨性关节炎模型组、透骨消痛颗粒干预组,术后第1、3、6、9、12、15周分批进行取材,采用石蜡切片苏木精-伊红染色、番红-固绿染色、AB-PAS染色和V-G染色,观察膝关节软骨和软骨下骨的变化,以及PCNA免疫组织化学染色观察软骨细胞和成骨细胞的增殖情况。结果:模型组关节软骨和软骨下骨发生骨性关节炎病理形态变化,软骨基质蛋白多糖尤其是酸性粘多糖减少,软骨和成骨细胞增殖早期较正常组活跃,晚期软骨细胞增殖能力下降;透骨消痛颗粒干预组与模型组相比,组织结构破坏和软骨下骨硬化明显减轻,蛋白多糖的含量增加,细胞增殖较稳定。结论:透骨消痛颗粒对实验性兔膝骨性关节炎软骨和软骨下骨病变均有一定的防治作用,这可能主要与其阻止或减缓OA过程中细胞外基质的降解作用有关。Objective:To investigate the effect of Chinese compound prescription Tougu Xiaotong Granule (TXG) on cartilage and subchondral bone of osteoarthritis rabbit models. Methods: Thirty-six New Zealand rabbits, half males and half females, were randomly divided into 3 groups: control group, osteoarthritis model group, and TXG treated group. Except control group, the model of knee osteoarthritis was established in other groups using modified Hulth method. The tissues were taken out in the 1st, 3rd, 6th, 9th, 12th and 15th week after operation and made into paraffin sections. HE staining, safranine-fast green staining, AB-PAS staining and VG staining were applied to observe the changes of cartilage and subchondral bone and immunohistochemical staining of PCNA was applied to observe the proliferation rate of chondrocytes and osteoblasts. Results:Osteoarthritis model group showed morphology of osteoarthritis. The proteoglycan, especially acid mucopolysaccharide and collagen in the cartilage matrix decreased. The ehondrocyte and osteoblast proliferated more actively than those of control group in early stage, yet the reproductive activity of chondrocyte attenuated in advanced stage. Compared with the model group, the histological structure of TXG treated group was significantly improved, the proteogly can increased, and the cell proliferation was more stable. Conclusion: TXG can prevent and cure the changes of cartilage and subchondral bone of osteoarthritis, which may be the result of its preventing the degradation of extracellular matrix.
