摘  要:目的:观察力竭游泳运动后大鼠伏核(NAc)内核转录因子-κB(NF-κB)和环腺苷酸(cAMP)的表达。方法:成年雄性SD大鼠随机分为对照组和力竭组。建立一周力竭游泳运动模型后,按0、4、8、12、16、20、24 h七个时间点,采用免疫印迹和免疫组织化学技术观察NF-κB在伏核内的表达变化,应用酶联免疫吸附技术检测伏核内cAMP的含量变化,并与对照组进行比较。结果:对照组伏核内NF-κB的表达较少,cAMP含量较低,力竭组NF-κB的表达明显增多,cAMP含量也明显增加,两者均于8 h达到较高水平,随后维持在较高水平,于16 h后开始回落,24 h已明显下降。结论:力竭游泳训练1周可以使大鼠伏核内NF-κB与cAMP得到有效的活化,且两者的表达变化具有明显相同时效性,提示它们可能具有协同效应。Objective:To observe the expression of NF-κB and cAMP in nucleus accumbens(NAc) of adult rats after exhausted swimming.Methods:Adult male SD rats were divided into the control group and exhausted group.Taking 0,4,8,12,16,20 and 24 hours after exhausted swimming for a week,Western Blot and immunohistochemical methods were used to detected the change of NF-κB in NAc,ELISA was used to examine the expressional change of cAMP.All results were combined with control group.Results:The expression of NF-κB and cAMP were extremely lower in control group,but increased in NAc after exercise,both reach higher levels at 8 h,then retainned higher levels for a moment,fell after 16 h and descended 24 h later dramatically.Conclusion:NF-κB and cAMP can be activated effectively in NAc after exhausted exercise,and the tendencys of change of them are similar.It is suggested that they have synergistic effect.
