摘  要:目的:为了比较大鼠足趾皮下注射福尔马林(formalin)和腹腔注射脂多糖(LPS)诱导下丘脑室旁核(PVN)内c-Fos蛋白表达的差异。方法:本实验采用免疫组织化学ABC双重细胞染色法,观测formalin和LPS应激后下丘脑室旁核大细胞部(mPVN)和小细胞部(pPVN)内c-Fos蛋白和精氨酸加压素(AVP)的定位和分布情况。结果:(1)与空白对照组比较,LPS剂量依赖性地诱导mPVN和pPVN内c-Fos蛋白表达(P<0.05);(2)与空白对照组比较,formalin剂量依赖性地诱导pPVN内c-Fos蛋白表达(P<0.05),但mPVN内c-Fos蛋白无明显增加;(3)当两组模型大鼠pPVN内c-Fos蛋白表达无明显差异时,与formalin组比较,LPS组mPVN内c-Fos蛋白表达明显增多(P<0.05),两者mPVN内的AVP表达无明显差异。结论:结果提示formalin和LPS均可引起下丘脑PVN神经元产生特异性反应,但LPS诱发的c-Fos阳性神经元分布相对广泛,结果提示下丘脑神经元的激活与刺激方式、强度等相关,单一模式、单一指标来揭示和衡量其功能状态是不完善的。Objective:To compare the different expression of c-Fos protein in hypothalamic paraventracular nucleus(PVN) of rats induced by formalin and lipopolysaccharide(LPS) stimulus.Methods:Using the dual-labeled immunohistochemical method,the location,distribution and expression of c-Fos protein and arginine vasopressin(AVP) in the magnocellular division(mPVN) and parvocellular division(pPVN) induced by formalin and LPS injection were observed.Results:Compared with control group,the number of c-Fos immunopositive neurons induced by LPS increased significantly in mPVN and pPVN in a dose-dependent manner(P0.05).Formalin could induced the number of c-Fos in pPVN but not in mPVN in a dose-dependent manner compared with control group(P0.05).LPS could induce the number of c-Fos in mPVN signficiantly compared with formalin group(P0.05),but c-Fos expression had no difference in pPVN.The neurons in PVN could respond specifically to stress induced by formalin and LPS,but the distribution of c-Fos induced by LPS is more widespread.Conclusion:The results indicate that it is less persuasive to choose the sole stimulus and the sole parameter to disclose and evaluate the activating properties of hypothalamic neurons which are related with the intensity and patterns of stimulus.
