摘 要:【目的】观察补肾壮骨冲剂对去卵巢骨质疏松大鼠骨组织形态学的影响。【方法】选用SD大鼠随机分为5组:假手术组,模型组,阳性对照组(仙灵骨葆胶囊,剂量为1.1 g.kg-1.d-1),补肾壮骨冲剂高、低剂量组(中药高、低剂量组,剂量分别为2.5、0.83 g.kg-1.d-1),每组雌雄大鼠各12只。各组在施行去势手术2周后给药,连续13周检测各组大鼠的胫骨近端骨小梁面积百分比、厚度、数目,骨小梁分离度。【结果】模型组雌性大鼠的胫骨近端骨小梁面积百分比、厚度、数目均显著减小(P<0.01),骨小梁分离度显著增加(P<0.01);雄性大鼠骨小梁面积百分比、数目均显著减小(P<0.01),骨小梁分离度显著增加(P<0.01),骨小梁厚度无显著改变(P>0.05)。中药高剂量组雌性大鼠骨小梁数目显著增加(P<0.05),骨小梁分离度显著减少(P<0.05),雄性大鼠骨组织形态指标均无显著改变(P>0.05)。中药低剂量组和阳性对照组雌雄大鼠骨组织形态指标与模型组比较差异均无显著性意义(P>0.05)。【结论】高剂量补肾壮骨冲剂能对抗雌性大鼠去势后所引起的胫骨近端骨小梁面积百分比与数目减少以及分离度增加,对雄性大鼠去势后所引起的骨质疏松无明显改善。Objective To investigate the effect of Bushen Zhuanggu Granules (BZG) on bone histomorphology in ovariectomized and castrated rats with osteoporosis. Methods SD rats were randomized into 5 groups, i. e. , pseudo-operation group, model group, positive control group (Xianling Gubao Granules 1.1 g·kg^-1·d^-1 ) , and high-dose and low-dose BZG groups (2.50 and 0. 83 g ·kg^-1·d^-1, respectively). There were 12 male and 12 female rats in each group. Rat models of osteoporosis were induced by removal of bilateral ovaries or testicles. Two weeks after operation, the rats were given the corresponding medicine. After medication for 13 weeks, rats were executed for the detection of the trabecula area percentage ( ThAt% ) , trabecula thickness (TbTh) , trabecula number (TbN) and trabecula separation (TbSp) in the proximal tibia. Results Compared with the pseudooperation group, the TbAr%, TbTh and TbN in female rats were significantly decreased ( P 〈 0. 01 ), and TbSp was remarkably increased (P 〈0.01 ) in the model group; the TbAr% and TbN in male rats were decreased (P 〈0.01), TbSp was remarkably increased (P 〈0. 01), and TbTh remained unchanged in the model group. In high-dose BZG group, the TbN was increased and TbSp was decreased in female rats (P 〈 0.05 compared with the model group). However, the effect on the histomorphological feature of male rats was not obvious in high-dose BZG group (P 〉 0.05 ). The parameters of trabecula in male and female rats of low-dose BZG group and positive control group did not differ from those in the model group (P 〉 0. 05 ). Conclusion High-dose BZG can improve the TbAr% , TbN and TbSp in ovariectomized osteoporotic rats, but has no effect on castrated osteoporotic rats.
