摘 要:目的通过探讨克疣灵对尖锐湿疣(CA)病人NK细胞活性,凋亡调控蛋白表达水平的影响作用以阐明克疣灵的正向免疫调节作用。方法抽取105例男性CA病人静脉血,采用自然沉降法获取白细胞、Ficoll-Urog分层液离心处理获取非黏性淋巴细胞、Percoll非连续梯度密度离心处理获取NK细胞,并进行NK细胞与不同剂量的克疣灵体外共同培养,采用放射免疫法;培养48 h后按不同的E/T测定NK细胞活性,培养12、24、36 h后测定NK细胞的增殖率,采用ELISA法于培养48、96 h后测定NK细胞凋亡调控蛋白sFas及NK细胞表面活性因子sCD226的表达水平。结果克疣灵高剂量组在培养12、24、36、48 h后能显著改善NK细胞的增殖率与活性,培养48 h后NK细胞凋亡调控蛋白sFas表达水平显著下调。NK细胞表面活性因子sCD226表达水平显著下调,NK细胞表面活性因子sCD226表达水平显著提高,上述指标与IL-15组比较差异无显著性(P>0.05),与克疣灵低剂量组、空白对照组比较有显著性差异(P<0.05或<0.01或<0.001)。结论高剂量克疣灵能显著改善NK细胞的功能,增强NK细胞的活性,降低NK细胞对凋亡的易感性,具有较强的正向免疫调节作用。Objective To clarify the positive adjustment effect by the experimental study of effect of Keyouling on the activation of NK cell.Methods After isolated the NK cell from the peripheral blood of 20 male CA patients by Ficoll-Hypaque centrifugation,the NK cell was cultured with different dosage of Keyouling.To determine the Proliferation and activation of NK cell with different E/T by means of 3H-TdR isotope release after cultured 12,24,36,48 hours.Results The Proliferation and the activation of NK cell had been significantly reinforced by the high dosage of Keyouling after cultured 12、24、36、48 hours.There were no significant difference of all the above detail compared with the group of IL-15(P0.05),but there were significant difference compared with the group(P0.05,P0.01 or P0.001).Conclusions The high dosage of Keyouling could reinforce the activation and proliferation of NK cell.It had significant positive immune adjustment effect.
