摘  要:目的探讨大鼠全脑缺血缺氧损伤后血液中S-100蛋白(S-100)、磷酸肌酸激酶脑型同工酶(CK-BB)、神经元特异性烯醇化酶(NSE)的水平变化。方法采用成年SD大鼠全脑缺血缺氧损伤模型:戊巴比妥腹腔注射麻醉后,用4.0双线结扎右侧颈总动脉,恢复4小时后,在37℃恒温低氧箱内,给予吸入8%氧气和92%氮气混合气体2小时。用放射免疫法和琼脂糖凝胶电泳法对对照组、缺氧缺血后6、12、24、48、72小时进行动态观测全脑缺血后血液中S-100、CK-BB、NSE水平的变化。结果在全脑缺血后,与对照组比较,血液和脑脊液中S-100、CK-BB、NSE在24~48小时都有明显增加(P<0.05)。结论在全脑缺血损伤时血液和脑脊液中S-100、CK-BB、NSE水平都在早期明显升高,提示这三种指标可作为评价大鼠全脑缺血缺氧损伤的早期诊断指标。Objective To analyse the changes of S-100 protein (S-100), creatine phosphokinase BB isoenzyme (CK-BB), and neuron-specific enolase (NSE) levels in blood after hypoxic-ischemic brain damage in rats. Methods The model ofhypoxic-ischemic brain damage in postnatal 180 days SD rats. The model was established by ligation of the right common carotid arteries for 4 hrs in rats, and then expo- sure of the rats to 8% of oxygen and 92% of nitrogen for 2 hrs. The blood samples were taken from 90 rats at the sequential time points of 6, 12, 24, 48, 72 hours after the model was established. Twenty-four normal rats were used as control. The dynamic changes of S-100, CK-BB and NSE were detected. Results All three biochemical markers significantly increased in blood and CSF of experimental rats when compared to controls. The peak level of S-100, CK-BB and NSE occured at 12 hrs after hypoxic-ischemic injury in blood (P 〈0.05). Conclusion The most obvious changes of S-100, CK-BB, NSE in hypoxic-ischemic injury rats were seen at 12 - 24 hours, the three biochemical markers might be helpful in the early diagnosis of hypoxic-ischemic cerebral injury.
