克毒宁胶囊对吗啡依赖猴的作用研究

摘　　要：目的:评价克毒宁胶囊对Mor(吗啡)依赖性猴的脱毒治疗作用。方法:采用剂量递增法建立猴Mor依赖性模型,进行猴自然戒断治疗试验,以停Mor前1 d(第90 d)的体重为基值,计算出戒断前后每天猴体重、体温变化百分率。计算各组戒断症状分值。结果:在猴自然戒断治疗试验中,克毒宁0.125、0.5和1.0 g/kg(1.07、4.28、8.55 g生药/kg)组戒断症状分值分别在戒断d1-4、d1-3、d1-2显著低于Mor模型组(P<0.01或P<0.05),其作用与可乐定相当。0.5 g/kg(中剂量)克毒宁在戒断d2显著抑制了戒断后体温下降(P<0.05)。克毒宁和可乐定组戒断后的体重下降百分率与Mor组比较无显著性差异((P>0.05)。结论:克毒宁胶囊对吗啡依赖性动物戒断后所产生的戒断综合征具有明显的治疗作用,其作用机理有待于进一步研究。Objective: To evaluate the detoxification effect of Keduning capsule on morphine-dependent monkeys.Methods: The morphine-dependent monkey models were established by the dose-increasing method to conduct monkey natural withdrawal tests by taking the body weight of 1 d(the first 90 d) before stopping the mor as the base value and calculate the body weight of the monkey per day before and after the withdrawal and the percentage of body temperature changes as well as their withdrawal symptom scores of each group.Results: In the treatment of the natural withdrawal test in the monkeys,the withdrawal symptom scores of Keduning capsule 0.125,0.5 and 1.0 g/kg(1.07,4.28,8.55g crude drug/kg) were d1-4,d1-3 and d1-2,significantly lower than that of the mor model group(P<0.01 or P<0.05),and its role was equal to clonidine.Keduning capsule 0.5 g/kg(middle dose) after d2 significantly inhibited the body temperature decrease(P<0.05) after the withdrawal.Comapared with the morphine group,there was no significant difference(P> 0.05) in the percentage of the body weight loss of Keduning and clonidine groups after the withdrawal.Conclusion: Keduning capsule shows a significance curative effect on morphine-dependent animals after the withdrawal,and its mechanism needs further study.
