摘 要:目的观察芪参活血颗粒对内毒素血症大鼠动脉收缩活性的影响,并初步探讨其可能的机制。方法将wistar大鼠分为空白对照组、模型组、芪参活血颗粒治疗组、芪参活血颗粒预防组。制备离体胸主动脉环,应用血管张力检测技术检测各组血管环对KCl的反应性并绘制剂量反应曲线。结果与空白对照组相比,内毒素血症大鼠胸主动脉血管环最大收缩反应明显降低(P<0.05);与内毒素组相比,芪参活血颗粒预防组和治疗组血管环收缩反应性明显增强;L-NAME处理后,模型组动脉环对KCl的最大收缩能力明显升高,芪参活血颗粒预防组及治疗组升高程度介于两组之间。结论内毒素血症时血管NO过量生成,导致血管的持续舒张状态和对收缩物质的低反应性;芪参活血颗粒能够下调iNOS的表达,减少舒血管物质NO的合成来提高血管对收缩物质的反应性。Objective:To observe the effect and the mechanism of Qishenhuoxue Granule on artery contractility changes of rats with endotoxaemia.Methods: Wistar rats were divided into 4 groups randomly including the control group,the LPS group,the treatment with LPS combined with Qishenhuoxue Granule group and the prevention with LPS combined with Qishenhuoxue Granule group.After the thoracic aorta rings were established,their responses to the KCl were detected by the angiotasis testing technology.And then,these results were drew dose-response curve.Results: Compared with the control group,the maximum contraction responses of the LPS group to KCl were reduced significantly.Compared with the LPS group,these response were increased obviously in the prevention group and the treatment group.After pretreatment by L-NAME,the maximum contraction responses of the LPS group to KCl were raised remarkably.Meanwhile,improvements in the prevention group and the treatment group were between the model group and the LPS group.Conclusion: The excessive production of NO level in endotoxmia can reduce the continued vessel diastole and the low vasoconstriction to vasoconstrictors.The Qishenhuoxue Granule can increase the response of the vessel to the vasoconstrictor by decreasing the iNOS expression and the NO synthesis.
