摘 要:目的 探讨中国南海角果木二萜成分Tagalsin的抗肿瘤作用。方法建立小鼠Lewis肺癌和H2肝癌实体瘤模型及H22肝癌腹水瘤模型。随机分为角果木二萜Tagalsin高、中、低剂量组(20、10、5mg/kg)、空白对照组、环磷酰胺(CTX)组。建模后24h开始给药,角果木二萜Tagalsin高、中、低剂量组连续灌胃给予不同浓度Tagalsin7d,空白对照组给予同体积生理盐水,CTX组连续腹腔注射CTX(10mg/kg)7d。末次给药24h后处死动物,计算抑瘤率、脏器指数、生命延长率,流式细胞术检测凋亡率。结果角果木二萜Tagalsin在5—20mg/kg内,对小鼠实体瘤Lewis和H22的抑瘤率分别为12.62%-43.69%和24.49%-48.30%。Tagalsin可明显延长Hz腹水瘤小鼠生存时间;降低Lewis肺癌及H。肝癌小鼠胸腺指数,升高肝脏指数;肿瘤细胞的凋亡率随着角果木二萜Tagalsin剂量的增加而增高(P〈0.05或〈0.01)。结论角果木二萜Tagalsin对小鼠Lewis和H22移植瘤的生长具有明显的抑制作用,其机制可能与其诱导细胞凋亡有关。Objective To study the anti-tumor effect of biterpenes tagalsin of cefiops tagal collected from the South China Sea. Methods The subcutaneously transplanted solid tumor models with Lewis and hepatoma-22 and the ascitic tumor models with hepatoma-22 were established. All the models were divided randomly into 5 groups : 5, 10 and 20 mg/ kg biterpenes tagalsin groups, saline and CTX ( 10 mg/kg), each of which was composed of 10 mice. The biterpenes tagalsin were given by intragastric administration, the CTX by intraperitoneal injection for 7 days, same volume of saline was given to the control group. The mice were killed 24 hours after the last medication, the the tumor inhibition rate, indexes of organ, increase in life span, apoptosis rate were calculated. Results With the dosage of 5-20 mg/kg (biterpenes Tagal- sin), the tumor inhibition rate in lewis model by 12.62%- 43.69%, and by 24.49%-48.30% in the hepatoma-22 model; slgnillcanlly prolonged the life span of mice in the ascitic tumor models with hepatoma-22 ; the thymus index increased and the hepatic index droped in both solid tumor models ; the apoptosis rate of the two kind of tumor cell correlated positively with the dosage of cembrane-type diterpenes( P 〈 0.05 or 〈 0.01 ). Condusions The biterpones tagalsin has significant anti-tumor effect by inducing apoptosis of tumor cells.
