摘  要:目的探讨肝硬化大鼠肝组织中组织胺H2受体基因的mRNA表达情况。方法逆转录聚合酶链反应方法从Wistar大鼠脑组织中合成H2受体的cDNA片段,克隆入载体中测序。用同位素标记cDNA片段后用于Northern印迹杂交。采用Northern印迹杂交法检测了10例肝硬化Wistar大鼠及10例正常对照组大鼠肝组织的H2受体mRNA。结果正常及肝硬化大鼠肝组织中均有H2受体的mRNA表达,且肝硬化组的表达量显著低于正常对照组。结论肝硬化时肝组织H2受体基因的转录减少是造成肝硬化时H2受体蛋白减少的重要原因,在门静脉高压症的形成和维持中起重要作用。Objective\ To investigate mRNA expression of histamine H 2 receptor (H 2R) in hepatic tissues of cirrhotic rats with portal hypertension. Methods\ Reverse transcription and polymerase chain reaction was used to synthesize cDNA of H 2R gene from Wistar rats' brain tissues and then cDNA was cloned into vector to be sequenced. The H 2R cDNA was labeled with isotope α 32 P and used as special probe in Northern blot hybridization. Then, mRNA expression of H 2R in hepatic tissues of 10 cirrhotic rats with portal hypertension and 10 normal control rats was detected by using Northern blot hybridization. Results\ mRNA expression of H 2R in hepatic tissues of both normal and cirrhotic rats was found, but in cirrhotic rats was significantly lower than that of normal rats ( P <0.01).Conclusion\ The decrease of H 2R gene transcription is an important cause of the decrease of H 2R proteins in the hepatic tissues of cirrhotic rats with portal hypertension, which might play an important role in the pathogenesis and maintenance of portal hypertension.
