摘 要:目的探索丝裂霉素C对含重复序列质粒的大肠杆菌致死作用的规律。方法采用均匀设计试验方法设计丝裂霉素C的处理时间和药物浓度,统计存活菌数,计算菌体致死率。结果得到了丝裂霉素C的浓度和作用时间与菌体致死率间的线性回归方程,建立丝裂霉素C作用的响应面。结论丝裂霉素C的浓度和作用时间与菌体致死率呈线性正相关。OBJECTIVE To explore the lethality law of E.coli containing plasmid pLC(GAA)42 treated by Mitomycin C.METHODS U*6(64) uniform experiment method was used to investigate the acting time and concentration of Mitomycin C and bacteria count was obtained by bacterial colony counter,then the lethality rate were counted.RESULTS The regression equation between lethality rate and concentration,and time of Mitomycin C was obtained and the correlation coefficient was 0.907.On the basis of the regression equation,response surface of mitomycin C treating E.coli DH5α was established.CONCLUSION There exists positive correlation between concentration,acting time of Mitomycin C and lethality rate of E.coli containing plasmid pLC(GAA)42.
