HPLC同时测定腰痛宁贴片中的士的宁和马钱子碱

摘　　要：目的采用RP-HPLC法同时测定腰痛宁贴片中士的宁和马钱子碱的含量。方法采用Inertsil C8色谱柱(250 mm×4.6 mm,5μm),流动相为甲醇-水-冰醋酸(100∶480∶35,三乙胺调pH3.2),流速1.0 mL.min-1,检测波长254 nm,柱温30℃,进样量10μL。结果士的宁和马钱子碱的线性范围分别为2.0～10.0μg(r=0.9998)、1.8～9.0μg(r=0.9997),平均回收率分别为98.9%(RSD=1.19%)、100.7%(RSD=1.49%),3批腰痛宁贴片中士的宁和马钱子碱的平均含量分别为5.15、4.03 mg。结论所建方法准确可靠,可用于腰痛宁贴片中士的宁和马钱子碱的含量测定。OBJECTIVE To establish an RP-HPLC method for simultaneous determination of strychnine and brucine in Yaotongning patches.METHODS The samples were separated on a Inertsil C8（250 mm × 4.6 mm,5 μm）at 30 ℃and detected at 254 nm.The mobile phase was consisted of methanol-water-acetic acid（100：480：35,adjusted to pH3.2 with triethylamine） with flow rate of 1.0 mL·min-1,and the injection volume of samples was 10 μL.RESULTS The results indicated that the two compounds were basically isolated.The linear ranges of strychnine and brucine were 2.0-10.0 μg（r=0.9998）,1.8-9.0 μg（r=0.9997）,respectively.The average recoveries were 98.9%（RSD=1.19%）,100.7%（RSD=1.49%）,respectively.The average contents of strychnine and brucine in a piece of Yaotongning patches were 5.15 mg and 4.03 mg,respectively.CONCLUSION The method is sensitive and accurate,and can be used to control the quality of Yaotongning patches.
