摘  要:目的观察鞘内注射神经生长因子(NGF)及抗神经生长因子抗体(anti-NGF)对骨癌痛大鼠疼痛行为学的影响,探讨NGF在骨癌痛中可能的作用。方法建立大鼠胫骨骨癌痛模型,鞘内置管,并分别注入生理盐水(cancer组)、NGF(cancer+NGF组)或anti-NGF(cancer+anti-NGF组),同时设立假手术组(sham组)。在不同时点观察疼痛行为学变化。结果与sham组比较,cancer各组大鼠体质量减轻;且cancer+anti-NGF组大鼠体质量比cancer组大鼠增加。与sham组比较,cancer组大鼠自发缩足次数增多,缩爪潜伏期(PWL)缩短,缩爪阈值(PWT)降低;与cancer组比较,cancer+anti-NGF组大鼠缩足次数明显减少,PWL延长,PWT增高;而cancer+NGF组大鼠与cancer组比较,缩足次数增多,PWL缩短,PWT降低。结论 NGF可加剧大鼠骨癌痛,而anti-NGF可显著减弱骨癌痛大鼠的痛敏。Objective To observe the effect of nerve growth factor(NGF)and anti-NGF injected intrathecally on pain behavior in rat model of bone cancer pain.Methods The rat model of bone cancer pain was established,and randomly assigned to receive intrathecal injection of normal saline(cancer group),NGF(cancer+NGF group),and anti-NGF(cancer+anti-NGF group).The changes in mechanical and thermal hyperalgesia were assessed at different time points after inoculation of Walker 256 tumor cell.Results Compared with shamoperated group,the body weight of rats in other 3 groups significantly decreased(P 〈 0.05);and the body weight of rats in cancer+anti-NGF group was significantly higher than that in cancer group(P 〈 0.05).Compared with sham-operated group,more paw withdrawal responses,reduced paw withdrawal latency(PWL),and decreased paw withdrawal threshold(PWT)were observed in cancer group.Compared with cancer group,the number of paw withdrawal response decreased,and PWL and PWT increased in cancer+anti-NGF group;but in cancer+ NGF group the number of paw withdrawal response increased,and PWL and PWT decreased.Conclusion NGF could deteriorate cancerinduced bone pain in rats,while anti-NGF could ameliorate the mechanical and thermal hyperalgesia.
