摘 要:目的探讨孤养和慢性不可预知的温和应激(CUMS)对抑郁模型大鼠海马星形胶质细胞内胶质纤维酸性蛋白(GFAP)和c-fos基因表达的影响。方法选取无特定病原体(SPF)级雄性W istar大鼠30只,随机分为对照组和模型组,应用孤养和CUMS建立抑郁模型,刺激时间8周,免疫组化法检测海马GFAP和c-fos基因的表达。结果模型组大鼠海马区GFAP表达增强,阳性细胞数增加,与对照组比较,差异有统计学意义(P<0.05);而c-fos随着应激时间的延长,表达减弱。结论孤养和CUMS可诱发抑郁,通过神经胶质细胞内GFAP和c-fos表达的变化,进而影响机体的行为及学习认知功能。Objective To discuss the expressions of glial fibrillary acidic protein(GFAP) in hippocampus and c-fos of rats with stimulation of raising separately and chronic unpredictable mild stress(CUMS).Methods 30 SPF male Wistar rats were randomly divided into 2 groups,a control group and a raising separately and CUMS model group.The model group was treated with 8 weeks of random stimulus.GFAP protein in hippocampus and c-fos was detected by immunohistochemistry.Results The expression of GFAP in model group was increased(P〈0.05).The expression of c-fos was weakened with the extension of stress.Conclusion Raising separately and CUMS could induce depression.Through the expression of GFAP and c-fos in astrocytes,behavior and cognitive learning could be affected.
