摘 要:【目的】研究青蒿琥酯诱导K562细胞凋亡过程中bcl-2表达的改变。【方法】培养K562细胞,倒置光显微镜和荧光显微镜观察青蒿琥酯处理的K562细胞形态,RT-PCR检测bcl-2的表达。【结果】K562细胞经青蒿琥酯处理48h后,倒置光显微镜:细胞出现不同程度的皱缩,核分裂相减少,细胞密度下降,细胞出现发泡现象,细胞内可见多个折光性较强的圆形小泡样结构。荧光显微镜:染色质高度浓缩、边缘化,凝集成明亮的团块。RT-PCR显示青蒿琥酯处理的K562细胞的bcl-2的表达下调。【结论】青蒿琥酯抑制K562细胞生长的机制可能与bcl-2基因的表达下调有关。【Objective】 To study the changes of expression of bcl-2 in the course of apoptosis of K562 cells induced by artesunate.【Methoods】 K562 cells were treated with artesunate of different concentration for 24h.The morphogical changes of K562 cells were observed with inverted microscope and Hoechst33342/PI staining.Expression of bcl-2 was determined by RT-PCR.【Results】 After treated with artesunate,K562 cells shrunk to certain degree,density of cells decreased and with vesicular form,division of necleus decreased.Many refractory oval vesicular formed structure could be found in the cells.Apoptotic changes as Karyopyknosis and conglomeration were found by Hoechst33342/PI staining RT-PCR showed down-regulation of bcl-2 expression.【Conclusion】 The mechanism of inhibiting K562 cell line by artesunate may be related with the down-regulation of bcl-2 expression.
