摘 要:【目的】探讨构建小鼠L1210白血病微小残留病模型所需注射的化疗药物的剂量。【方法】将不同数量L1210白血病细胞接种清洁级近交系DBA/2小鼠(n=10),观察接种的白血病细胞数量与小鼠存活时间之间的关系;将一定数量的L1210白血病细胞接种DBA/2小鼠(n=10),接种后第3天静脉注射不同剂量的环磷酰胺(CTX),观察药物注射剂量与小鼠存活时间之间的关系;将接种L1210白血病细胞数量与DBA小鼠存活时间关系图和CTX注射剂量与白血病小鼠存活时间关系图相结合,确定构建小鼠L1210白血病微小残留病模型所需注射的化疗药物的剂量。【结果】小鼠存活时间随接种细胞数量增加而缩短,接种500个白血病细胞小鼠的存活时间约为28 d;白血病小鼠存活时间随药物剂量增加而延长,存活时间约28 d时,需要静脉注射CTX 125 mg/kg。【结论】构建小鼠L1210白血病微小残留病模型,可于DBA/2小鼠接种L1210白血病细胞1×106个/只后第3天,给予CTX 125 mg/kg静脉注射。【Objective】 To investigate the dose of chemotherapeutic drugs in construction of minimal residual leukemia model of L1210 cells in mouse.【Methods】 Different amount of L1210 cells were inoculated into depuratory inbred DBA/2 mice(n=10) and the relationship of the amount of inoculated cells and the survival time of mouse was investigated.L1210 cells of 1×106 per mouse were inoculated into DBA/2 mice.On the third day,cyclophosphamide(CTX) of different dose was infused into these mice through vena caudalis and the relationship of the dose of CTX and the survival time of mouse was investigated.Results of the above two steps were combined to decide the dose of CTX in construction of minimal residual leukemia model of L1210 cells in mouse.【Results】 The survival time of mouse was shortened with the increase of amount of inoculated cells and the survival time was concluded about 28 days while 500 cells were inoculated into mice.The survival time of leukemia mice was lengthened with the increase of dose of infused CTX and the dose of infused CTX was about 125 mg/kg while the survival time was 28 days.【Conclusion】 CTX of 125 mg/kg can be infused into leukemia mice through vena caudalis 3 days after inoculation of L1210 cells of 1×106 per mouse into depuratory inbred DBA/2 mice to construct the minimal residual leukemia model of L1210 cells in mouse.
