通脉大生片对雷公藤致卵巢损伤大鼠性激素与抑制素的影响

摘　　要：目的:探讨补肾法中药复方干预性治疗的作用机理,为中医药调节女性生殖轴的研究提供一个新的符合中医证候特点的病证结合动物模型。方法:选雌性育龄期SD大鼠行阴道脱落细胞学涂片,筛选出动情周期正常者为受试动物,除正常组外,其余大鼠予雷公藤多苷致动情周期紊乱模型。雷公藤模型大鼠随机分为即空白组、倍美力组、中药高、中、低剂量组5组,分别予倍美力和通脉大生片不同剂量治疗7天,空白组同期给予等量生理盐水,正常组正常喂养。各组均于治疗前后采血,称重法测算卵巢指数;放射免疫法检测血清雌二醇、孕酮;酶联免疫吸附法检测抑制素B水平;免疫组化法检测卵巢雌激素受体(ER)、孕酮受体(PR)。结果:①予雷公藤多苷40 mg/kg.d连续灌胃10周,可造成育龄期大鼠动情周期紊乱或消失,卵巢指数下降,血中E2、INHB水平明显降低;卵巢组织中ER表达明显降低;②运用通脉大生片干预性治疗后,大鼠动情周期恢复至正常范围,卵巢指数升高;血清E2、INHB水平及卵巢组织中ER表达均明显升高;与空白组相比均有显著统计学意义。结论:雷公藤多苷可造成大鼠卵泡发育障碍和卵巢功能低下,补肾气益精血的中药复方"通脉大生片"能显著提高模型大鼠血清雌二醇和抑制素B水平及卵巢中ER表达,从而有效地提高雌激素的生物效应和卵巢储备功能。Objective：Referring to the establishing method of mouse model by our previous research,the aim of this study was to build a model of rattus norregicus＇ ovarian follicle suppression caused by Tripterygium Glycosides,and probe the action mechanisms of the interfering therapy using Bushen herbal compound.Thus,to provide the research on the regulation of female reproductive system by TCM with a novel combination of disease and syndrome animal model corresponding to TCM syndrome character.Methods： Give vaginal cytology assay to all the female SD rats of reproductive age,and filter the laboratory rats which have normal estrous cycle.Take 6 rats randomly as normal control group,and give the others intragastric administration with tripterygium glycosides 40 mg/kg/day for 10 weeks,then,observe the rats＇ estrous cycle,whose disorders infer to the success of creating the model.Divide the rats randomly into five groups,6 for each,including the blank group,the Premarin group and herbal different-dosage group.Feed the animal subjects with Premarin and Tong-Mai-Da-Sheng Tablet in different dosages separately for seven days,and control group with normal saline.Take blood before and after the treatment respectively.After seven days,kill the rats.Measure the ovarian index by tissue-weight-assay,check out E2,P with RIA,and Inhibin B with ELISA.Detect ER,PR with IHC.Results：（1） Intragastric administration with tripterygium glycosides 40 mg/kg/day for 10 weeks can create the model of rattus norregicus＇ ovarian suppression,which manifests as follows： disorders of estrous cycle,decreasing of ovarian index and E2,ER,INHB in blood,decreasing of ER in ovarium,increasing of Endostatin and blood vessel wall thickness/area ratio.（2） Tong-Mai-Da-Sheng Tablet has sound effects in regulating the estrous cycle,increasing the ovarian index,E2,ER,INHB in blood.These data were all significantly different from the blank control group.Conclusion：The tripterygium glycosides can create the model of rattus norregicus＇ ovarian f
