摘 要:目的:探索复合牛骨形态发生蛋白(bBMP)骨软骨生物修复材料的细胞相容性及其对于兔关节骨软骨缺损的修复效果。方法:将bBMP经处理制成复合bBMP骨软骨生物修复材料(A材料),并将。体外培养扩增兔骨髓基质干细胞(MSC)传至第三代后接种于A材料中,体外培养2周后扫描电镜(SEM)观察材料的细胞相容性。将A材料植入兔关节骨软骨缺损,12w后取材观察其修复效果。结果:A材料孔隙结构规则,孔径约100~200μm,孔隙内部均匀附着bBMP,与兔MSC共培养显示了良好的细胞相容性。在兔关节骨软骨缺损修复中,A材料12周实现了对关节骨软骨较好的修复效果。结论:复合bBMP骨软骨生物修复材料在保持理想孔隙结构和细胞相容性的同时,其骨软骨缺损修复能力明显提高。Objective: To explore biocompatibility of morphogenetic protein(bBMP) that was a osteochondral repairing biomaterial incorporated with bovine bone and its repaired effect on rabbit osteochondral defect. Methods: bBMP was processed and formed compound osteoehondral repairing biomaterial with Bbmp(A materials), medullary stem celI(MSC) of rabbit had a serial subcultivation to third generation and were inoculated into A materials, MSC had a culture in vitro and biocompatibility were observed by SEM examine after 2 weeks,Then implant the biomaterials into rabbits' osteochondral defects, 12 weeks later, the reparied effect tissues were observed microscopically. Results:A materials had a regular pore structure and pore size was 100-200μm,and the bBMP film was evenly adhered onto the biomaterial. The biomaterial showed very good biocompatibility when co-culturing with rabbit MSCs. In the osteochondral repairing experiment, the biomaterial loaded with bBMP repaired osteochondral in 12 weeks. Conclusion:Compound osteochondral repairing biomaterial with Bbmp presented very good structure and biocompatibility while its bioactivity on osteochondral repairing was greatly improved.
