摘  要:目的:研究燕窝蛋白质在模拟胃肠环境中的降解情况。方法:制备燕窝水提物,分别与等体积的人工胃/肠液混合,37℃消化,按时间梯度0、1、5、10、15、30、60 min准确计时并终止反应,Bradford法检测上清液的蛋白质浓度,SDS-PAGE凝胶电泳方法检测燕窝蛋白质的降解情况。结果:燕窝总蛋白经胃蛋白酶或胰蛋白酶消化后,大部分降解到40 kD以下,部分中高分子量的蛋白质不降解,而且在70 kD左右新增一条蛋白条带。结论:部分中高分子量的燕窝蛋白质具有抗消化性,推测可能以抗原的形式直接作用于肠上皮细胞启动肠道免疫系统发挥疗效。Objective:To study the digestion of Bird′s nest extract(BNE) in simulated gastric fluid(SGF) and simulated intestinal fluid(SIF).Methods:BNE were prepared and digested at 37 ℃ in SGF/SIF.At intervals of 0,1,5,10,15,30,60 min,the samples were taken out.Protein contents were determined by the method of Bradford,and digestion of BNE in SGF/SIF was observed according to the results of polyacrylamide gel electrophoresis(SDS-PAGE).Results:On SDS-PAGE,the crude BNE and the BNE in SGF/SIF gave different patterns,but all being rich in high molecular weight protein.After digestion in vitro,most of protein degraded to 40 kD below,but part of high molecular weight protein was stable,and a protein of 70 kD added.Conclusion:Most protein degraded to polypeptide,but there was anti-digest protein in the BNE,all being the high molecular weight protein.This suggested that the active component from BNE was probably exerted directly on human body.
