摘  要:目的:研究环丙沙星体外诱导铜绿假单胞菌耐药的外排泵机制。方法:多步诱导法对5株铜绿假单胞菌进行体外诱导耐药实验,应用外排泵抑制剂(CCCP)选择出外排泵阳性的铜绿假单胞菌菌株,将上述铜绿假单胞菌诱导前后的菌株分别行增菌培养,提取外膜蛋白,行蛋白电泳,紫外凝胶成像后分析诱导前后外膜蛋白中OprM量的变化来研究环丙沙星体外诱导对铜绿假单胞菌外排泵的影响。结果:诱导后的5株铜绿假单胞菌中外排泵阳性的为3株,蛋白电泳显示诱导后铜绿假单胞菌外膜中外排泵蛋白OprM含量明显增多。结论:环丙沙星体外诱导铜绿假单胞菌耐药的机制之一是增加细菌外膜外排蛋白的含量。Objective:To investigate the induced resistance to Ciprofloxacin and the mechanism of efflux pumps in Pseudomonas aeruginosa.Methods:Indueed resistance to Ciprofloxacin was performed by multiple-step method in 5 strains of Pseudomonas aeruginosa. To select positive efflux pump CIP-resisitance Pseudomonas aeruginosa by CCCETaking the Pseudomonas aeruginosa that before and after induced resistance to Ciprofloxacin,to culture and extract outer membrane protein(Omp),then to go on sodium dodecyl sulfate-polyacrylamide gel electrohoresis (SDS- PAGE) and to photoed in the ultraviolet gelatin machine to calculate the concentration of OprM,to deduce the mechanism of effinx pumps in Pseudomonas aeruginosa by induced resistance to Ciprofloxacin.Results:The result of sodium dodecyl sulfate- polyacrylamide gel elcctrohoresis (SDS- PAGE) shows that the concentration of OprM of Pseudomonas aeruginosa outer membrane increase obviously by induced resistance to Ciprofloxacin.Conelusion:The mechanism of Pseudomonas aeruginosa induced resistance to Ciprofloxacin is increase the concentration of effinx pumps.
