摘 要:目的 建立一种压电石英晶体传感器检测血浆凝血因子Ⅷ的方法.方法 采用AT切向、基频10MHz的银膜石英晶体作为压电元件;观察血浆凝集过程中晶体振荡频率的变化,确定血浆凝集反应的起点和终点.结果 ①将乏Ⅷ因子血浆、活化部分凝血活酶时间试剂与待测血浆混匀,加入检测池;待频率稳定后,加入CaCl2于检测池,观察反应频率,在凝集反应结束时的频率变化由黏度、密度、质量、晶体表面及凝块的厚度、凝块性质的变化而决定,加样后频率下降的起点为血浆凝集反应的起点T1,频率上升、下降,稳定的起始点为反应的终点T2,计算两点的时间即为反应时间Tc=T2-T1,查标准曲线得血浆凝血因子Ⅷ的活性.②用该方法检测50例临床标本所得平均血浆凝血因子Ⅷ活性为107.4%,STAGO磁珠检测法检测50例临床标本所得平均血浆凝血因子Ⅷ活性为102.0%,该方法的检测结果与STAGO磁珠检测法的相关性好(r=0.948).结论 压电石英晶体传感器检测血浆凝血因子Ⅷ是一种实时在线检测的方法,在线观测凝集反应的终点,反应的起点和终点的判别方便、可靠、准确,且操作简单、快速、成本低廉,是一种敏感度高、重复性好的方法,并可用于临床检测.Objective To establish method of using piezoelectric quartz crystal sensor to detect blood plasma factor Ⅷ. Methods A silver wafer with a basis resonance frequency 10 mHz AT-cut quartz crystal was used as the piezoelectric component ,and the oscillation frequency shift of the quartz crystal during the blood coagulation was monitored. The start and end point of the blood coagulation reaction was decided. Results (1)To mix the lack Ⅷ plasma,activated partial thromboplastin time reagents under test plasma by adding testing pool and was frequency stabilized by adding CaCl2 on the detection of pool ,observed the reaction of the frequency,in the agglutination frequency change by the end of the viscosity, density,quality,crystal surface and the thickness of clot,clot the changing nature of the decision to increase the frequency decrease after sample plasma agglutination reaction from the starting point for the T1 ,the frequency up,down,stable starting point for the reaction to the end of T2,two points of time should be calculated response time Te=T2-T1 ,checked the standard curve to get the activity of plasma coagulation factor Ⅷ. (2)The method detected 50 cases of clinical specimens from the average activity of plasma coagulation factor Ⅷ 107.4% and STAGO magnetic bead detection assay of clinical specimens from 50 patients with average plasma eoagulation factor Ⅷ activity was 102.0 %,and the detecting results by this method was correlated with the results by STAGO magnetic bread method greatly (r= 0. 948). Conclusion Using piezoelectric quartz crystal sensor to detect blood plasma factor Ⅷ is an on-line detecting method. It can monitor the startpoint and end-point of the coagulation reaction in real time. It is convenient ,dependable ,and accurate to find the start-point and end-point of the coagulation reaction. It is a sensible and repeatable method which is simple,quick and cheap. It can be used in clinical detection.
