窒息新生猪脑皮层细胞凋亡与淋巴细胞凋亡的相关性研究

摘　　要：目的　探讨新生儿窒息时淋巴细胞调亡与脑细胞凋亡是否具相关性,以期寻求围生窒息时临床客观判定脑细胞凋亡的指标。方法　制作新生猪HIE模型,用琼脂电泳及流式细胞术对HIE新生猪及正常对照新生猪淋巴细胞及脑细胞凋亡进行定性、定量分析。结果　HIE新生猪(n= 15)淋巴细胞及脑细胞凋亡率均显著高于正常对照组(n= 12)。F= 8.67　P< 0.01,窒息新生猪淋巴细胞凋亡与脑细胞凋亡呈显著正相关,r= 0.4007　P= 0.038。Objective To determine the relation between apoptosis of cerebral cortical cells and apoptosis of lymphocytes in neonatal pigs following perinatal asphyxia. Methods A flow cytometry and gelelectrophoresis were used to quantitate and qualitatively analyse the apoptosis after neonatal pig HIE model was prepared. Results Apoptotic rate of lymphocytes in vitro (26.72%± 16.01% )and of cerebral cortical cells(27.50%±16.12%)in vivo 72 hr later after asphyxia in experimental group( n =15)were significantly higher than those(3.95%±3.90%,3.23%±2.93%)in control group( n=12),F =8.67 P <0.01.The apoptotic rate of lymphocytes,was significantly correlative to the apoptotic ratio of cerebral cortical cells, r=0.4007 P=0.038. Conclusion The apoptotic rate of lymphocytes in vitro may be a useful clinic index to estimate the apoptosis of cerebral cortical cells in vivo in neonates following perinatal asphyxia.
