摘 要:目的观察丁苯酞对AD大鼠模型的学习记忆能力及海马Aβ、NR2b表达的影响。方法采用大鼠海马立体定向注射凝聚态Aβ1-40诱导AD模型。采用Morris水迷宫评价大鼠学习记忆能力,采用免疫组织化学染色法测定海马区Aβ、NR2b蛋白的表达。结果丁苯酞低剂量组和高剂量组能明显减少海马Aβ的沉积,有效促进海马CA1区NR2b的表达(P<0.01);对AD大鼠模型的学习、空间记忆能力有明显的改善(P<0.01);而高剂量组和低剂量组对AD大鼠模型的改善作用没有明显差异。结论 AD大鼠模型学习记忆的改变与海马Aβ、NR2b变化相关,丁苯酞可能通过减少海马Aβ的沉积、促进CA1区NR2b的表达从而改善AD大鼠模型的学习和空间记忆能力。Objective To observe the effects of butylphthalide on learning,memory and expressions of hippocampus of Aβ and NR2b in AD rats models.Methods Aggregated Aβ1-40 was injected into the hippocampus of rats by stereotaxic coordinates method,to induced Alzheimer's models.Morris water maze test was used to determine the ability of learning and memory,Aβ and NR2b in hippocampus was measured by immunohistochemical staining.Results Low and high dose group of Butylphthalide could significantly reduce Aβ deposition in hippocampus and promote the expression of NR2b in hippocampus CA1 area effectively(P0.01).Learning and spatial memory of AD rats models improved significantly(P0.01).But there was no significant difference between high-dose group and low-dose group for improving effect of these aspects.Conclusion The change of Aβ and NR2b in hippocampus was related to the learning and memory of AD rats models.Butylphthalide can improve the learning and memory ability of AD rats may by reducing the deposition of Aβ in hippocampus and promoting the expression of NR2b in CA1 area.
