摘  要:目的:研究早期生长反应基因-1(Egr-1)对血管平滑肌细胞(VSMC)增殖、凋亡和表型转化的影响。方法:实验分空白对照组和Egr-1组。应用MTT法研究不同浓度和不同作用时间组Egr-1对VSMC增殖的影响;应用透射电镜方法、原位DNA片段末端标记(TUNEL)方法,研究不同作用时间下Egr-1对VSMC表型转化与凋亡的影响。结果:MTT法检测不同浓度Egr-1组与对照组间比较OD值具有显著性差异(P<0.05)。浓度40μg/mL时促VSMC增殖效应最显著。在最适浓度下(40μg/mL)Egr-1促VSMC增殖作用于24h达到高峰。Egr-1抑制VSMC凋亡的作用在12h时最明显。对VSMC凋亡的影响在一定时间范围内呈负相关。Egr-1作用24h时促VSMC向合成表型转化的作用最显著。结论:Egr-1对VSMC的促增殖作用在一定范围内与浓度和作用时间呈正相关。Egr-1一定程度具有抑制VSMC凋亡的作用并促进VSMC由收缩型向合成型转化。Objective:To investigate the effects of early growth respons gene-1(Egr-1) on the proliferation,apoptosis and phenotype transformation of vascular smooth muscle cell(VSMC).Methods:The VSMCs of the rat thoracic aorta were subcultured by the method of adherence.The third to eighth generation of cells was selected for the experiment.The experiment consisted of the control group and Egr-1 group.We applied the MTT method respectively to study the effects on the proliferation of VSMC by Egr-1 at different concentrations and the different effect time.The method of transmission electron microscope and TUNEL were used to study the effect of Egr-1 with different effect time on phenotype transformation and apoptosis of VSMC.Results:There were significant differences in OD values tested by MTT assay at different concentrations of Egr-1 group compared with the control group,(P0.05).Egr-1 showed the most significant effect on promoting VSMC proliferation when the concentration was at 40μg/mL.Atr the optimal concentration(40μg/mL),the peak of VSMC proliferation promoted by Egr-1 was at 24h.The most obvious action of inhibiting VSMC apoptosis by Egr-1 was at 12h.The effect of Egr-1 on VSMC apoptosis was negatively correlated within a certain time.When it was at 24h,Egr-1 played the most significant role in promoting converting to synthetic phenotype of VSMC.Conclusion:Promotion proliferation action on VSMC by Egr-1 is positively correlated with the concentration and action time within a certain range.To some extent,Egr-1 has an effect on inhibiting VSMC apoptosis and promoting VSMC converting from contractile type to synthetic type.
