摘 要:对来自不同地区的15份苦楝个体用RAPD技术分析其遗传多样性,20个随机引物共扩增出193条重复性好、清晰的位点,其中多态位点共计189个,占97.9%.聚类分析表明:云南保山与四川绵阳种源间的遗传距离最大(0.936),福建永泰与福建建瓯之间的遗传距离最小(0.186).应用UPGMA聚类分析法构建了DNA分子系统树图,为基于DNA分子水平深入探讨苦楝不同种源间的亲缘关系和系统位置奠定了良好的基础,在散生乡土树种遗传多样性研究方面进行了开拓性和有意义的探索.Genetic diversity of 15 samples for Melia.azedarach Linn from different provenances were analyzed by RAPD technique.193 clear and repeat sites were totally detected by 20 primers.The polymorphic sites were total 189,polymorphic degree was up to 97.9%.The results of cluster analysis indicated that the maximum genetic distance(0.936) between Baoshan Yuannan and Mianyang Sichuan and the minimum genetic distance(0.186) was between Yongtai Fujian and Jianou Fujian.A dendrogram by unweighted pair groupmethod with arithmetic mean algorithm(UPGMA) analysis was obtained.Its result was basically identical with morphologic taxonomy,which was laid sound foundation for probing into the interspecies relatives and the location of the system of different provenances of Melia.Azedarach Linn based on the DNA molecular level.And pioneering and meaningful explorations were carried on scattered indigenous tree genetic diversity.
