大鼠腹主动脉平滑肌细胞的培养与鉴定

摘　　要：目的探讨一种简单、经济获取腹主动脉血管平滑肌细胞的方法,为相关研究提供实验材料。方法应用改良组织贴块法进行原代培养,胰酶消化传代,运用自然纯化法进行细胞纯化,并使用相差显微镜和SP试剂盒对细胞进行形态学观察以及免疫组化鉴定。结果 90%的组织块接种存活,培养细胞呈典型的"谷峰状"生长,免疫荧光化学技术检测显示胞浆内α平滑肌肌动蛋白阳性表达。结论本法简便可靠,短期内可获大量高纯度、功能良好的血管平滑肌细胞。Objective A simple and economical way to obtain abdominal aorta vascular smooth muscle cells（VSMCs） was investigated for the related researches.Methods The primary and transfer culture were done by modified tissue-piece inoculation and trypsin digestion respectively.The cells were purified by natural purification.The cultured cells were identified by morphology and immunohistochemistry with phase contrast microscope and SP kit.Results 90% inoculated tissue pieces survived.The cultured cells showed the typical ＂peak and valley＂ morphological appearance.Immunofluorescence detection staining with mAb against mouse SM-α-actin demonstrated these cells were positive.Conclusion A large number of high-purity and well-functioning VSMCs were obtained by this simple and reliable method.
