摘 要:目的建立Beagle犬海水浸泡致低体温及合并腹部开放性创伤动物模型;研究并比较左氧氟沙星在海水浸泡致低体温及合并腹部开放性创伤Beagle犬动物模型及正常Beagle犬的药物动力学。方法 12只Beagle犬随机分为3组,每组4只。组1为正常对照组,组2为低体温模型组,组3为腹部开放性创伤合并低体温模型组。给药方案为:左氧氟沙星25 mg/kg,静脉输注,60m in输注完毕。SPSS程序进行统计学处理。结果组1动物给药前后72 h体温正常,为(37.20±0.27)℃;组2和组3海水浸泡后体温明显下降,最低分别为(30.00±1.12)℃和(29.8±1.03)℃。组2、组3与组1比较,消除半衰期、药物峰浓度以及药时曲线下面积明显增加,清除率有所降低。相同时间点血药浓度比较,组2、组3均高于组1。组2与组3间药物动力学参数及药物浓度均无差异。结论在72 h内海水浸泡致低体温状况可导致左氧氟沙星药物动力学特性变化,腹部开放性创伤的影响不显著。Objective To investigate pharmacokinetics of levofloxacin in beagle dogs under hypothermia with open abdominal wound by artificial seawater immersion and normal body temperature.Methods Twelve healthy beagle dogs were randomly divided into three groups(4 each):control group(group 1),hypothermia model group(group 2)and open abdominal wound and hypothermia model group(group 3).The beagle dogs in group 2 and group3 were immersed into artificial seawater for 90 min before being administered to prepare beagle dog hypothermia models.All the animals were administered with a single dose of levofloxacin(25 mg/kg) by infusion into the saphenous vein.Results The temperature of animals in group 1 predose and at 72 h postdose was normal(37.20±0.27)℃,but those in group 2 and group 3 were significantly decreased,the lowest values being(30.00±1.12)℃ and(29.8±1.03)℃ after immersion process,respectively.In comparison to those of group 1,t1/2β,Cmax,C12,C24 and AUC of animal in group 2 and group 3 were both significantly increased,respectively.ClS of group 2 and group 3 were both decreased compared with that of group 1.There was no significant difference in the main pharmacokinetic parameters between group 2 and group 3.Conclusion Hypothermia is an important factor that leads to difference in pharmacokinetics of levofloxacin in beagle dogs within 72 h while open abdominal wound had no effect.
