高效液相色谱法测定葛黄颗粒中葛根素的含量

摘　　要：目的建立葛黄颗粒中葛根含量的高效液相色谱测定方法。方法采用Shimpack VP-ODS色谱柱：（4.6 mm×250 mm,5μm）;流动相：乙腈-水-冰醋酸（14∶85∶1）;流速：1.0 ml/min;检测波长：254 nm;柱温：室温;峰面积外标法定量。结果葛根素在0.02260.226μg范围内线性关系良好（r=0.9998）,平均回收率为100.35%,RSD为1.01%（n=6）。结论该方法测定样品分离度佳,灵敏度高,重复性好,结果准确可靠。Objective To establish an high performance liquid chromatography method for determination of puerarin in Gehuang granule.Methods Shimpack VP-ODS（4.6 mm×250 mm,5 μm）was used with acetonitrile-water-acetic acid glacial（14∶85∶1） as the mobile phase.The flow rate was 1.0 ml/min,detected at 254 nm and room temperature.The external standard method was adopted for the assay.Results The injection quantity of Puerarin presented a good linear range of 0.0226-0.226 μg（r=0.9998）.The average recovery was 100.35%,RSD=1.01%（n=6）.Conclusion The method is good in separability,reproducibility and sensitivity.
