摘 要:目的:建立肺炎克雷伯菌的基因扩增多态性DNA分型方法,了解产ESBLs肺炎克雷伯菌院内感染的分子流行情况。方法:收集临床各病区送检的产ESBLs肺炎克雷伯菌感染标本29份。采用随机扩增DNA多态性方法对产ES-BLs肺炎克雷伯菌进行基因分型。利用Phylip3.67聚类分析软件对RAPD扩增条带结果进行聚类分析。结果:RAPD扩增的指纹图谱清晰,带型稳定,多态性丰富。29株不同来源的菌株可大致分为三种亲缘关系。结论:随机扩增多态性DNA技术可以用于产ESBLs肺炎克雷伯菌基因多态性分型研究;产ESBLs肺炎克雷伯菌基因多态性分布可能与菌株的来源有关。Objective:To genotype clinical Klebsiella pneumoniae isolates by randomly amplified polymorphic DNA(RAPD) method,study molecular epidemiology of extended-spectrum beta-lactamases(ESBLs) producing Klebsiella pneumoniae infection.Methods: 29 ESBLs Klebsiella pneumoniae isolates with corresponding patients information.All the isolates were genotyped by RAPD.Cluster analysis was performed using Neighbor Joining method by Phylip3.67 soft.Results: Primer was suitable for fingerprinting analysis and was found to generate the reproducible fingerprints,yielding well-resolved banding patterns.Generally,the isolates can be divided into three clusters.Conclusion: RAPD fingerprinting can be used to genotype Klebsiella pneumoniae;the RAPD patterns of ESBLs Klebsiella pneumoniae may be related to origins of fungi;it seems the RAPD patterns also related to drug resistance.
