摘 要:本试验应用聚丙烯酰胺垂直板电泳分离、分析了旋毛虫抗原的组分。电泳后经考马斯亮蓝染色的旋毛虫幼虫抗原呈现20 条蛋白质色带;经Schiff 试剂染色呈现6 条红色的糖蛋白色带。将电泳后的凝胶均匀分割Ag1 、Ag2 、Ag3 、和Ag44 个组分。分别测定其蛋白质浓度为1237μg、2254μg 、2636μg、2604μg 。糖蛋白区带成分属于Ag1 和Ag4 。经ELISA 和免疫电泳检测,Ag1 组分的抗原活性最强,其次Ag2 ,Ag3 抗原活性微弱,Ag4 无抗原活性。通过试验证实Ag1 组分具有最强的抗原活性,敏感度高,而且属于大分子糖蛋白成分,它是作为检测旋毛虫病的首选最佳抗原成分。This test used PAGE to analyse the antigene components of the muscle larvae of Trichinella Spiralis After electrophoresis and staining with Coomassie Brilliant Blue G250,the antigene components of the T Spiralis were found to be composed of 20 protein bands By using the glycoprotein specific Schiff′s reagent,six of them were shown to be red glycoprotein band Then the gel was cut evenly into 4 parts,Ag 1,Ag 2,Ag 3,and Ag 4 The protein concentration of them were 12 37μg,22 54μg,26 36μg,and 26 04μg respectively The glycoprotein bands belonged to Ag 1 and Ag 4 separately Tested by ELISA and immunoelectrophoresis,Ag 1 was found to have the greatest activity,Ag 2 was the second,and the antigenicity of Ag 3 was quite weak,yet Ag 4 was shown to have no activity at all As a glycoprotein component,Ag 1 was proved to have the greatest activity and sensitivity,thus it′s the best choice for the testing of T Spiralis
