长吻鮠血细胞的显微、亚显微结构及细胞化学研究

摘　　要：光镜下长吻鮠外周血中可区分出红细胞、中性粒细胞、单核细胞、淋巴细胞和血栓细胞,未见嗜酸性粒细胞和嗜碱性粒细胞,其中,红细胞数量最多,其次是血栓细胞和淋巴细胞,单核细胞和中性粒细胞数量最少;同时还发现幼稚红细胞、直接分裂的红细胞.电镜下,个别红细胞可见到少量线粒体;中性粒细胞表面有短而钝的伪足样突起,颗粒有3种类型;单核细胞内有较多的溶酶体样颗粒;小淋巴细胞有细长的伪足样突起,大淋巴细胞内质网丰富.细胞化学染色显示:所有白细胞PAS反应呈阳性;SBB染色只有嗜中性粒细胞呈阳性;单核细胞及嗜中性粒细胞和个别淋巴细胞中含有ACP,只有粒细胞含有POX;单核细胞、嗜中性粒细胞均含有ANAE,部分淋巴细胞含有ANAE.Examination under a light microscope of the Wright-Giemsa stained smears of blood cells of Leiocassis longirostris Günther has revealed the following major cell types： erythrocyte,neutrophilic granulocyte,monocyte,lymphocyte and thrombocyte.Eosinophilic granulocytes and basophilic granulocytes haven＇t been seen in the peripheral blood smears.Among these cells erythrocyte is the most abundant.Thrombocyte and the lymphocyte are the most abundant leucocytes.Monocyte is as much as neutrophilic granulocyte and is the least encountered leucocyte.In addition,the immature and dividing,dissolved are also found.Dividing erythrocyte（bilobed erythrocyte） is a form of senescence.The bilobed morphology marks the process of apoptosis.Thrombocyte is always irregularly round to oval,tadpole-shaped and spindle-shaped,and the dividing thrombocytes are also found.Observed under the transmission electron microscope,erythrocyte,which is shuttle-shaped,has rarely organelles and some mitochondria can be found in it occasionally.The small lymphocyte is characterized by its long pseudopodia and large lymphocyte processes abundant of endoplasmic reticulum.Some granules,like lysosome,are seen in monocyte.Neutrophilic granulocyte is recognized by its many specific granules,and these granules have three types.Neutrophilic granulocyte has pseudopodia,which,however,are shorter than those of small lymphocyte.The erythrocytes are PAS negative while the leucocytes positive.Only neutrophilic granulocytes are SBB positive.Acid phoshpatase exists in monocytes and neutrophilic granulocytes and very few lymphocytes have acid phoshpatase.No alkaline phoshpatase exists in all types of blood cells.Peroxidase exists only in neutrophilic granulocytes.α-naphthyl acetate esterase is found in neutrophilic granulocytes and monocytes.Only part of lymphocytes have α-naphthyl acetate esterase.
