摘 要:采用PCR-SSCP技术研究了不同窖龄浓香型白酒窖池古菌群落的变化规律,结果发现:①所有窖泥样品均出现7~12条较清晰的条带,其中第19,26号条带在所有样品中均有检出,且优势度较高;②随着窖龄的增加,窖池不同位置样品的第19号条带优势度基本上都呈先增加后降低的趋势,第26号条带优势度基本上都呈先降低后增加的趋势;③所有窖泥样品平均生物多样性指数都在1.78~2.26之间,随着窖龄的增加,窖壁上层与窖底样品的多样性指数呈增加趋势,窖壁下层样品呈先降低后增加的趋势;④不同窖龄窖池SSCP图谱相似性指数较低,在0.17~0.43之间,而同一窖池不同位置样品的相似性指数较高,在0.42~0.95之间.The changes of Archaea community in Luzhou-flavor liquor pits of different pit ages were analyzed using PCR-SSCP technique.Seven to twelve distinct bands were shown in all pit mud samples,and bands NO.19 and NO.26 were detected in all samples,with relatively high dominance.With increasing pit age,the dominance of band NO.19 in all samples from different pit spots first increased and then decreased,while the dominance of band NO.26 first decreased and then increased.The mean biodiversity index of all samples ranged from 1.78 to 2.26.With increasing pit age,the biodiversity index of the samples from upper pit wall and pit bottom showed an increasing trend;the biodiversity index of the samples from lower pit wall tended to decrease first and then increase.The similarity index of SSCP pattern of the samples from different pit ages was rather low,between 0.17and 0.43,while the similarity index of the samples from different positions of the same pit was higher,between 0.42 and 0.95.
