摘 要:目的:观察α-硫辛酸干预对INS-1细胞的作用及相关机制。方法:先用α-硫辛酸(200μmol/L)加入INS-1细胞中一起培养6 d,再应用不同浓度的葡萄糖作用于INS-1细胞48 h,抽取上清液测定MDA含量,并检测GSIS值,同时应用RT-PCR的方法检测UCP 2基因的表达,并与不加α-硫辛酸组互相比较。结果:(1)在相同糖浓度下,LA干预组与未加LA干预组相比,MDA含量升高,GSIS值降低,各浓度组相比均具有显著性差异;(2)在相同糖浓度下,-αLA干预组与未加-αLA干预组相比,UCP2mRNA表达增强且差异均有显著性。结论:-αLA为一个促氧化剂,能引起MDA含量增加,UCP2mRNA表达增强,GSIS值下降,说明-αLA在较长时间作用于胰岛细胞时起到促氧化的作用。Objective:To explore the effect of α-lipoic acid on INS-1 cells and the related mechanisms.Methods:α-lipoic acid 200 μmol/L was added into INS-1 cells for 6 days,glucose at different concentrations were applied in INS-1 cells for 48 hours.MDA were detected by spectrophotometer,then insulin secretion was measured by ELESA with high-glucose stimulation and UCP-2 gene expression was detected by RT-PCR.And all indexes were compared with these of none α-lipoic acid intervention group.Results:1 With the same glucose concentration,MDA in INS-1 cells with α-LA intervention was higher than that without intervention,and the GSIS value was reduced with significant differences among groups.UCP2 mRNA expression in intervention group increased,and there were significant differences among different groups.Conclusions:α-LA,as a promoting oxidants,can promot the expression of UCP2 mRNA and decline the glucose-stimulated insulin secretion,while increased MDA in INS-1 cells shows that α-LA can promote oxidation when affecting the islet cells during a long period of time.
