摘 要:目的:检测血管内皮细胞生长因子(VEGF)在骨关节炎(OA)软骨中的mRNA表达。方法:16只大耳白兔随机分为实验组和对照组,实验组行单膝前交叉韧带切断术,对照组不做处理,术后9周处死动物,采用反转录-聚合酶链反应(RT-PCR)及实时荧光定量PCR方法检测软骨中VEGF的mRNA表达。结果:实验组软骨中VEGF的mRNA表达水平较对照组显著升高(P<0.05)。结论:骨关节炎软骨中VEGF的mRNA高表达,VEGF在骨关节炎软骨退变中发挥着重要作用。Objective: To investigate the expression of vascular endothelial growth factor (VEGF) mRNA in osteoarthritic cartilage. Methods: Sixteen white rabbits were randomly divided into 2 groups. Experimental group underwent unilateral anterior cruciate ligament transection (ACLT) to establish the models of osteoarthritis(OA). Animals in control group were not treated. All rabbits were killed 9 weeks following surgery, and the mRNA expression of VEGF in cartilages was analyzed using reverse transcription-polymerase chain reaction (RT-PCR) and real time PCR methods. Results: The level of VEGF mRNA expression in cartilages in the experimental group was higher than that in the control group (P0.05). Conclusion: VEGF mRNA expression was enhanced in the cartilage of the OA model. The result suggested that VEGF may be involved in cartilage degeneration of OA.
