摘　　要：目的用高脂饲料喂养2型糖尿病MKR鼠建立2型糖尿病并发非酒精性脂肪肝(NAFLD)动物模型。方法用高脂饲料诱发MKR鼠形成NAFLD模型,观察其肝脏形态、糖耐量、肝功能和血脂变化。结果高脂饲料诱发MKR鼠形成NAFLD模型肝细胞呈小泡性脂变,细胞内有大小不一的脂滴存在;糖耐量异常,异常程度高于MKR组。与C57野生组比较,MKR组小鼠肝质量、肝指数和血脂LDL-C升高差异无统计学意义(P>0.05),肝功能指标ALT、AST和血脂TG、TCHO、HDL-C升高差异有统计学意义(P<0.05或0.01);MKR高脂组小鼠肝质量、肝指数、肝功能指标(ALT、AST)和血脂(TCHO、HDL-C、LDL-C)进一步升高,高于MKR组(P<0.05或0.01),而TG下降,低于MKR组(P<0.01),高于C57野生组小鼠(P<0.05)。结论高脂饲料喂养MKR鼠可以建立稳定的、与临床2型糖尿病并发NAFLD一致的动物模型。Objective To establish type 2 diabetes combined with non-alcoholic fatty liver disease（NAFLD） model by feeding high fat diet to MKR mice.Methods The 8 week old MKR mice were fed high lipid diet for 8 weeks（H-MKR）,and the MKR mice（MKR） and C57 wild mice （C57） of the same age were served as the control group that were fed base diet.The pathological changes of liver,carbohydrate tolerance,blood-lipid and liver function were examined.Results Hepatocytes had fatty degeneration in H-MKR group and normal morphology in MKR and C57 groups.However,lipid droplets were seen in the hepatocytes of H-MKR.Compared with C57,carbohydrate tolerance in H-MKR and MKR were all abnormal,and that of H-MKR was worse.Compared with C57,liver weight,hepatosomatic index,LDL-C in MKR had no significant differences（P0.05）, otherwise ALT,AST,TG,TCHO and HDL-C had significant difference（P0.05 or 0.01）.Liver weight,hepatosomatic index,ALT,AST,TCHO,HDL-C and LDL-C in H-MKR increased further than those in MKR（P0.05 or 0.01）,but TG in H-MKR was lower than that in MKR（P0.01）,and higher than that in C57（P0.05）.Conclusion A stable NAFLD model which was in accordance with the clinical features of patients with type 2 diabetes and NAFLD was established.
