摘  要:以灭活的大肠杆菌免疫产蛋母鸡,应用酶联免疫吸附法(Enzyme-linked immunosor-bent assay,ELISA)测定鸡卵黄中免疫球蛋白(Immunoglobulin of egg yolk,IgY)的活性,并用水稀法、盐析与亲和层析结合的方法制备高效价卵黄抗体.结果显示灭活的大肠杆菌能诱导鸡产生高效价的抗体,效价高达1.6×105,经上述纯化技术路线,制备出了高纯度的IgY.卵黄是一种含量高、经济的特异性抗体源,为疾病的防治与检测提供了新的途径.Hens immunized with inactive E.coli,during immunization,the antibody immune activity and titer of anti E.coli immunoglobulin of egg yolk(IgY) were evaluated by enzyme-linked immunosorbent assay(ELISA),Anti E.coli IgY were isolated and purified by water dilution and salting out combined with affinity chromatography.Results show that the antigen of inactive E.coli strain was able to stimulate the hens to produce a large amount of high titer IgY.The highly purified anti E.coli IgY were obtained through the method mentioned above,and the ELISA titer of the purified IgY was higher than 1.6×105.The egg yolk of immunized chicken is a rich and inexpensive source of specific antibody which provides a new approach to the treatment and diagnosis of disease.
