摘  要:应用PCR-RFLP分子标记技术,研究了258只滩羊角蛋白KAP1.3基因的多态性及其与裘皮品质的相关性.结果表明,检测出A,B两个等位基因,基因频率分别为53%,47%;产生AA,AB,BB基因型,基因型频率分别为25.2%,55.8%,19.0%;BB基因型所对应的毛股弯曲数显著高于AA,AB基因型(P<0.05),AB,BB基因型在A+194 bp处碱基T转换为C,导致氨基酸由异亮氨酸变异为苏氨酸.可通过改变KAP1.3 BB基因型的频率来提高滩羊裘皮弯曲数目,也说明角蛋白KAP1.3基因有望成为提高裘皮弯曲数性状的分子标记辅助选择候选基因.The study was conducted to find the polymorphism of KAP1.3 gene and the correlation between KAP1.3 gene and fur quality characteristics of Ningxia tan sheep by PCR-RFLP.Alleles A and B were found in the present study,and their gene frequencies were 53% and 47%.Whilst the two alleles generated three different genotypes including AA,AB and BB with the genotype frequencies of 25.2%,55.8%,and 19.0%,respectively.The mean wool curvature marked with BB genotype was significantly higher than others(P〈0.05).In addition,the results of genetic sequencing showed that there was a nucleotide mutation(from T to C) at DNA A+194 of KAP1.3 gene in AB and BB genotype,and this mutation could result in amino acid change(from Ile to Thr).The results indicate that it is necessary to change KAP1.3 BB genotype frequency to improve the fur curvature and wool quality.KAP1.3 may therefore be expected to be molecular marker-assisted selection candidate genes which can increase the fur curvature.
