摘 要:目的查明广西汉坦病毒的基因型及其基因亚型。方法用夹夜法捕获宿主动物,用免疫吸附试验(ELISA)检测鼠肺标本,对汉坦病毒抗原阳性标本用RT-PCR法扩增,产物直接测序,拼接后获得全S基因序列,用生物软件进行核苷酸及氨基酸系列分析和病毒的系统进化分析。结果应用ELISA法检测鼠肺抗原792份,阳性6份,RT-PCR测定其中5毒株,基因型为汉城病毒,进一步的核苷酸序列分析均为S2亚型。结论广西汉坦病毒基因型为汉城型汉坦病毒,亚型为S2。Objective To investigate the genetypes and sub-types of Hantavirus in Guangxi province.Methods Host animals were collected and Hantavirus in the lung tissues were identified by Enzyme-Linked Immunosorbent Assay(ELISA).The complete S segments of positive samples were amplified with RT-PCR and sequenced.Phylogenetic analysis was used for genotyping.Results 792 specimens of lung tissues were detected by ELISA,and 6 were positive.Five of six positive specimens were detected by RT-PCR and sequenced,and all Hantaviruses were Seoul virus.Phylogenetic analysis of complete S segments showed that Seoul Hantaviruses in Guangxi province were the S2 sub-genotype,and were closely related with the strain from Zhejiang province.Conclusion The genotype of Hantaviruses in Guangxi province was SEO,and the sub-genotype was S2.
