摘  要:2010年6月以来,我国部分地区所饲养的种鸭和蛋鸭发生一种疾病,以突发产蛋急剧下降为特点,根据病变,暂将该病称为鸭出血性卵巢炎(duck hemorrhagic ovaritis,DHO)。从不同地区的发病鸭群采集34份样品,取15份样品进行病毒分离,获得10个鸡胚分离物和5个鸭胚分离物。RT-PCR检测结果表明,15个分离物均为禽流感阴性,14个分离物为新城疫阴性。选分离株YY5株进行了电镜观察和PCR排查,结果表明,该毒株的毒粒直径为40~50 nm,呈鸭瘟病毒、水禽细小病毒、鸭呼肠孤病毒、鸭甲肝病毒、鸭星状病毒、鸭圆环病毒、鸭冠状病毒、鸡传染性支气管炎病毒PCR阴性,但为黄病毒RT-PCR阳性。基于黄病毒NS1序列合成引物,用RT-PCR检测15株病毒分离物和其余19份临床样品,黄病毒阳性率为82.4%。用YY5株的221-nt NS1序列进行分析的结果显示,YY5株与黄病毒科黄病毒属恩塔亚病毒群(Ntaya virus group)的巴格扎病毒(Bagaza virus)具有相近的遗传进化关系,但遗传距离介于病毒种的水平。用一株鸭胚分离株感染91日龄临近开产的麻鸭和232日龄的产蛋麻鸭,可复制出卵泡膜出血的病变。结果表明,从DHO自然病例分离到一种新的黄病毒,暂称为鸭黄病毒(duck flavivirus,DFV),DFV可能与DHO有关。From June through November 2010,a disease characterized by a sudden onset was observed in many egg-laying and breeder duck farms in China.Based on the pathological changes,the disease was tentatively called duck hemorrhagic ovaritis(DHO).Thirty-four samples were collected from the diseased ducks in different regions and 15 of the samples were subjected to virus isolation by using SPF chicken and duck embryos.Fifteen isolates were obtained.RT-PCR results showed that all the 15 isolates were negative for avian influenza and 14 of the 15 isolates were negative for Newcastle disease.One strain,designated YY5,was selected for further investigation.Electron microscopic observation revealed that the virion in the isolate was 40 to 50 nm in diameter.PCR screening indicated that the isolate was negative for Anatid herpesvirus 1,duck parvovirus,egg-drop syndrom virus,duck reovirus,duck hepatitis A virus,duck astrovirus,duck circovirus,duck coronavirus and infectious bronchitis virus.However,the isolate was positive for flavivirus.RT-PCR detection based on the primers specific to flavivirus NS1 gene showed that the flavivirus was detected in 12/15 viral isolates and 16 of the remaining 19 samples.In total,the DFV positive rate was 82.4%.Sequence analysis based on a 221-nt NS1 sequence revealed that strain YY5 was most closely related to Bagaza virus(BAGV) in Ntaya virus group in the genus Flavivirus of the family Flaviviridae.Nevertheless,only 75% nucleotide identity was observed between YY5 and BAGV.Experimental infections with a duck-embryo-derived strain resulted in reproduction of the disease in 91-day-old and 232-day-old egg-laying ducks.The results demonstrated that a potential new flavivirus,designated duck flavivirus(DFV),was isolated from the DHO cases and that the DHO may be caused by DFV infection.
