摘 要:目的探讨异氟醚预处理在大鼠皮层神经元缺氧无糖(OGD)损伤的保护作用及其可能机制。方法建立原代培养的大鼠皮层神经元OGD损伤模型,采用MTT法观察异氟醚预处理对大鼠皮层神经元OGD损伤的保护作用,Westernblot法检测磷酸化ERK(pERK)和低氧诱导因子(HIF)-1α的蛋白表达水平。结果 OGD组神经元存活率显著低于其它四组(P<0.05)。IO组pERK、HIF-1α蛋白表达明显高于其它四组(P<0.05)。结论异氟醚预处理对大鼠原代皮层神经元OGD损伤具有明显的保护作用。其机制可能与异氟醚增加pERK表达,进而调节HIF-1α的表达有关。Objective To study the protective effect of isoflurane preconditioning on rat cortical neurons with oxygen-glucose deprivation (OGD) injury.Methods OGD injury was set up in primary rat cortical neurons. Protective effect of isoflurane preconditioning on rat cortical neurons with OGD was evaluated with MTT. Protein expression of pERK and HIF-1α were examined with Western blot. Results The level of hypoxia inducible factor (HIF-1α) in the rat cortical neurons with OGD injury increased after isoflurane preconditioning,which was partially abolished by ERK inhibitor PD98059. Conclusion Isoflurane preconditioning could protect the rat cortical neurons with OGD injury,possibly via upregulation of HIF-1α mediated by the increased pERK.
