摘 要:目的观察组胺对老龄大鼠肠道细菌易位的影响。方法将老龄大鼠随机分为对照组、肠道感染组和肠道感染+组胺组。除对照组外,其他大鼠均从肠道灌注3×1011/L标准大肠杆菌O75菌液0.4ml。菌液灌注后,肠道感染+组胺组立即给予小肠内100 ng/ml浓度的组胺,对照组和肠道感染组大鼠用同样方法向小肠内灌注等量生理盐水,4 h后在无菌条件下取出肝、肾、肠系膜淋巴结进行细菌培养。结果对照组大鼠肝、肾、肠系膜淋巴结平均细菌含量分别为1.02×105、0.62×105、0.38×105cfu/g;肠道感染组分别为8.35×105、7.16×105、5.68×105cfu/g,肠道感染+组胺组分别为5.25×105、4.06×105、3.58×105cfu/g。肠道感染+组胺组大鼠肝、肾、肠系膜淋巴结中的细菌含量明显低于肠道感染组(P<0.05)。结论小剂量组胺小肠内灌注有保护老龄大鼠肠黏膜屏障、阻止肠道细菌易位的作用。Objective To observe the effects of histamine on the translocation of intestinal bacteria by determining the bacteria content in the liver,kidney and mesenteric lymph nodes of aged rats.Methods 30 rats aged 24 months were randomly divided into control,intestinal infection and intestinal infection with histamine groups.Rats except those in control group were infused with standard escherichia coli O75 bacterium fluid(3×1011/L) 0.4 ml into the intestines through an infusion pump.After that,histamine solution(100 ng/ml) was immediately infused into the small intestine of the rats in intestinal infection with histamine group,at an infusion speed of 1.5 ml/h for 4 hours.The same volume of normal saline was given to the rats in control group and intestinal infection group.After 4 hours from the infusion,the liver,kidney and mesenteric lymph node of the rats were dissected out under aseptic conditions for cell culture.Results The average value of bacteria content in liver,kidney and mesenteric lymph node from the control group was 1.02×105,0.62×105,0.38×105 cfu/g,respectively.The value from the intestinal infection group was 8.35×105,7.16×105,5.68×105 cfu/g,respectively.And the value from the intestinal infection with histamine group was 5.25×105,4.06×105,3.58×105 cfu/g,respectively.The average bacteria content in liver,kidney and mesenteric lymph node of the intestinal infection with histamine group was obviously lower than that of the intestinal infection group(P0.05).Conclusions The perfusion of histamine at small dose has a role in protecting the intestinal mucosa barrier and preventing the translocation of intestinal bacteria in aged rats.
