摘 要:目的观察bio-oss载体对兔骨髓基质多潜能细胞贴附后的影响。方法体外培养骨髓基质多潜能细胞,进行成骨化的诱导,并进行前苏红钙化结节染色进行鉴定;诱导后的细胞滴加到bio-oss载体上,利用扫描电镜观察材料对细胞的形态学,利用MTT法观察材料对细胞增殖能力的影响。结果骨髓基质干细胞4~5 d可见少量的集落形成,14 d细胞基本铺满瓶底。第四代细胞在矿化液条件下培养2周左右,细胞形态稍变圆,茜素红染色结果显示强阳性。细胞滴加到载体材料后,可以很好的贴附于材料表面,增殖生长,材料对细胞增殖能力影响无差异。结论 bio-oss载体材料具有较好的生物相容性,可以作为骨组织工程的骨架材料。Objective: To observe bio-oss carrier on bone marrow stromal cells layering impact.Methods: After the culture of bone marrow stromal cells in vitro,ossification induction was made and former suhong calcification nodules dying was done for identification;and then induced cell dropped onto bio-oss carrier,morphology of cells was observed by scanning electron microscope,the cell proliferation ability was observed by the MTT.Results: Small colony formation of bone marrow stromal cells was visible in 4-5 d and bottle bottom was filled in 14d.Fourth-generation cells were mineralized fluids in 2 weeks and the bottle bottom was filled with cell morphology in slightly varying circle.Alizarin red staining results showed the strongly positive.Cells dropped onto the carrier material,and were very good layer to the material surface in multiplication,and there was no difference in cell proliferation capacity.Conclusion: As bone tissue engineering of reinforcing materials,Bio-oss carrier materials have good biocompatibility.
