肝细胞ABCB4基因与胆囊胆固醇结石关系的研究

摘　　要：目的探讨胆囊胆固醇结石的形成与肝脏组织内ABCB4基因表达的相关性。方法实验组选取14例胆囊胆固醇结石患者的肝组织标本,以10例非胆囊结石患者的肝组织标本为对照组。应用RT-PCR技术检测两组患者肝脏组织ABCB4基因的表达强度;应用免疫组化方法检测两组患者肝脏组织Pgp3蛋白的表达强度,所有检测结果行统计学分析。结果实验组和对照组ABCB4mRNA相对表达量分别为(0.91±0.06)和(1.15±0.04),两者比较差异有统计学意义(P<0.05),实验组Pgp3蛋白表达量(14.29%)明显低于对照组(80%)(P<0.05)。结论 ABCB4基因的表达降低使其编码的Pgp3蛋白表达降低,从而可能导致胆囊胆固醇结石的形成。Objective To explore the relationships between the expression of ABCB4 gene and the formation of cholesterol calculus. Methods Fourteen hepatic tissue specimens were taken from patients with cholesterol calculus.As comparison,ten were taken from patients without cholesterol calculus.The mRNA expressions of ABCB4 gene were determined by RT-PCR analysis,the expressions intensity of Pgp3 in hepatic tissue were determined by immunity histochemistry staining method respectively.Results The mRNA expressions of ABCB4 gene were significantly lower in patients with cholesterol calculus（0.91±0.06）than those without calculus（1.15±0.04） （P0.05）.The expressions of Pgp3 were significantly lower in patients with cholesterol calculus（14.29%）than those without calculus（80%） （P0.05）. Conclusion The decreased expressions of ABCB4 gene and Pgp3 may correlate with the formation of cholesterol calculus.
