摘 要:以幼嫩叶片、顶芽及茎段为外植体,研究不同外植体、消毒时间、抗氧化剂预处理、褐化剂以及不同培养基组合等对外植体褐变控制的效果。结果表明,嫩叶、顶芽及茎段的褐化率均随着0.1%升汞溶液消毒时间的延长而升高,而诱导率和污染率不断降低,最佳外植体为顶芽,消毒时间为10 min,诱导率达70.0%。采用3种抗氧化剂预处理20min,均可抑制顶芽褐化,强弱表现为PVP>Vc>Na2S2O3,Vc溶液可以加快侧芽的萌动及生长。3种抗褐化剂对茎段褐化均有不同程度的抑制效果,以0.3%AC抗褐化效果最佳,茎段未发生褐化,侧芽生长最好。在不同激素配比培养基中,随着6-BA和NAA浓度的提高,外植体发生褐化时间呈现提前的趋势,以MS+6-BA 2.0 mg/L+NAA 0.2 mg/L培养基中的外植体诱导率最高,为63.4%。此外,相对于基本培养基MS、1/2MS,在低无机盐离子浓度WPM基本培养基中,外植体褐化的时间有所推迟,可减缓外植体褐化程度。因此,适宜的外植体、消毒时间、抗氧化剂、抗褐化剂、激素组合配比和基本培养基,可以有效控制牛尾菜的褐化,提高诱导率。The present experiment was conducted to study the effects of different explants, sterilization time, antioxidant and anti-browning agents and culture media combinations on browning control of Smilax ripada A.DC explants. The young leaf, apical bud and stem of Smilax dpada A.DC were used as explants. The results showed that the browning rate of young leaf, apical bud and stem increased with the elongation of sterilization time in 0.1% mercury bichloride solution, while the induction and contamination rate were decreased. The best explant was found to be the apical bud, which showed 70.0% induction rate when sterilized for 10 min. All the three antioxidative agents were found to inhibit the browning of apical buds, when pretreated for 20 rain, of which the effects of polyvinyl pyrrolidone (PVP) was the best, followed by vitamine C (Vc)and Na2S203, the Vc solution was also found to promote the germination and growth of axillary buds. Three anti-browning agents showed varied effects on controlling the browning of stem, and the effects of 0.3% activated charcoal (AC) were found to be the best due to complete control of browning and well growth of axillary buds. An early browning was observed with increasing the concentrations of 6-BA and NAA in the media. However, the induction rate of explants was found the highest (63.4%) in MS medium supplemented with 6-BA (2.0 mg/L) and NAA (0.2 mg/L). Compared with the basic and 1/2MS medium, the browning of explants in low salt woody plant medium (WPM) was lesser and found to be delayed. The present studies revealed that suitable explants, proper sterilization time, use of antioxidant and anti-browning agents, and proper hormone combinations in basic medium could effectively control the browning of Smilax riparia A.DC explants and increase the induction rate.
